In the modern times, vesicular system is a well-known carrier for delivery of drugs. They can carry both lipophilic and hydrophilic Proniosomes are vital member novel drug carriers. The present study was aimed to develop prepare Metronidazole Benzoate loaded proniosomes with different ratios cholesterol non-ionic surfactants prolong release on topical administration. By slurry method, about fourteen formulations were prepared evaluated angle repose, content, particle size, hydration rate, encapsulation efficiency, in-vitro studies & ex-vivo permeation studies. optimized formulation developed into transdermal patch subjected various evaluation kinetic diffusion showed 80.71% over 12hrs fitted zero order non-fickian mechanism. It summed up, that F12 containing surfactant: as 1:1 best formulation. Incorporation lead reduced dose, improved bioavailability prolonged release. Further formulating in form allows controlled drug. UV spectrophotometric method determining benzoate 5% ethanol at 309nm. A regression coefficient value 0.9977 noticed. SEM image well FT-IR spectrum taken support same. Hence, we conclude proniosomal candidate metronidazole treatment systemic bacterial infections
